Project Title

Postdoctoral Researcher in Immunology : Mechanisms of hepatitis E virus immunopathogenesis in solid organ transplant recipients

Research Project

Hepatitis E virus genotype 3 (HEV) establishes mostly asymptomatic, self- limiting infection in immunocompetent individuals. However, in immunocompromised patients with solid organ transplant the infection progresses frequently to a chronic course. To date, the mechanisms involved in viral persistence remain to be discovered.

The overall objective of the current project is to investigate, at the cellular and molecular levels, the function of immune cells, particularly T cells, at the acute stage of the infection in order to unveil their role in viral persistence/clearance. To this end, cryopreserved PBMC and plasma samples (collected at the early and late phases of the infection) from infected and uninfected transplanted patients will be used. T cell subsets will be studied with embedded levels of analysis (phenotype, function, metabolism, gene expression) by flow cytometry and single cell Cite-seq. Additional metabolomics and proteomics studies will also be conducted on plasma samples.

Main Activities

· To be responsible for the development of the research project: plan experiments, analyze and interpret experimental data, report regularly to the supervisor and team members on scientific activities and maintain accurate and up-to-date records of all findings.

· To maintain an up-to-date awareness of relevant publications in the area of research.

· To participate in publications and funding request writing.

· To train and supervise junior members of the team.

Associated Activities

· To participate in the team project (experiments and analyses).

Scientific Environment

The Toulouse Institute for Infectious and Inflammatory diseases (INFINITy) provides a stimulating environment to perform state-of-the-art research in immunology and infectious diseases, with integrated on-site technological facilities. Research teams at the Center host international trainees of various nationalities, ranging from undergraduate students to postdocs. External seminars and lab meetings are held in English.

The host team, headed by Dr. Jabrane-Ferrat is currently composed of 9 members working collaboratively. The candidate will be supervised by the Dr. H. El Costa (PI). The research project will be in close collaboration with the French Nation Reference Center for entero-transmissible hepatitis E (Pr. J. Izopet and Dr. F. Abravanel).

Skills/Qualifications

· The applicant should have a strong expertise and proven research skills in T cells biology and immunology techniques such as multiparametric flow cytometry (FACS Symphony A5, Spectral Cytek Aurora) and metabolism analyses (Seahorse, Scenith).

· Proven practical experience in bioinformatics/Omics are mandatory.

· Experience using flow cytometry analysis software (OMIQ, FlowJo), statistical programs such as Prism and Microsoft office programs (Excel, PowerPoint, Word).

· Excellent communication skills, team spirit, capacity to work autonomously and deliver results on time.

Specific Requirements

PhD in immunology with 2 years of postdocoral experience.

Handling human samples in a 2/3 level containment laboratory.

Possible outside working hours.
